The Lexington Opera House originally opened in 1886 as the Grand Opera House. Throughout its history it has hosted a diverse range of performances including theatrical productions, vaudeville shows, concerts, and lectures. Significant renovations occurred in 1978 following a period of decline and near demolition; these restorations aimed to return the venue to its original grandeur while updating facilities. Further improvements were completed in 2006.

Today, the Lexington Opera House operates as a performing arts center owned by the City of Lexington and managed by Lexington Center Corporation.
